본 연구는 승마추출물(cimicifuga rhizome )을 이용하여 구강편평상피암(oral squamous cell carcinoma, SCC 25 ) 세포에서의 세포 사멸 유도 및 경로를 확인하기 위해 수행되었다. 본 연구에서는 SCC 25 세포에서 승마추출물에 대한 세포 생존력을 검출하기 위해 WST-1 분석을 사용하였다. 면역 형광 염색법 및 유세포 분석법은 구강암 세포의 세포 자멸사를 검출하기 위해 사용되었다. 그 후, 승마추출물이 세포 사멸과 관련하여 단백질(PARP-1, caspase 3 및 caspase 9)에 미치는 영향을 단백질 면역 분석법으로 확인하였다.
본 저자들은 승마 추출물이 SCC 25 세포에서 농도 의존적 방식으로 세포 생존력을 유의하게 억제한다는 것을 발견하였다. 또한 승마추출물은 sub G1을 증가시키고 cytochrome c와 AIF 위치를 변경하였다. 또한, caspase-3 신호 전달 경로가 승마추출물의 의해 유도된 세포 사멸에서 핵심적인 역할을 한다는 것을 관찰하였다. 본 연구 결과 승마추출물이 SCC 25 세포에서 caspase-3 신호 전달 경로를 조절하여 세포 사멸을 촉진하는 것으로 나타났다.
