High triglyceride (TG) levels are known to beassociated with complex diseases such as cardiovasculardisease. Here, we evaluated the effect of interactionsbetween genetic polymorphisms and environmental factorson plasma TG levels in a Korean aging cohort. Thirty-twosingle nucleotide polymorphism (SNP) markers located inseven genes (ADLH2, APOE, GCKR, MC4R, TCF7L2,GATA2, and HNF1A) for TG levels were genotyped in 714older Koreans (mean age, 68 years). We performed multiplelinear regression analyses for these SNP markersunder three genetic models (i.e. additive, dominant, andrecessive) and evaluated their associations with multipleenvironmental factors, including obesity, systolic bloodpressure, and drinking and smoking habits. We foundevidence for four SNP-smoking interactions. The CCgenotype of rs1260326 (GCKR) interacted with currentsmoking for lower TG levels (PG9E = 0.017). TG levelswere higher in smokers with the GG genotype ofrs2713604 (GATA2) than in those with the GA ? AAgenotypes (195.6 and 145.7 mg/dL, respectively,PG9E = 0.029). The GA ? AA genotypes of rs2713603(GATA2) were associated with elevated TG levels insmokers when compared to non-smokers carrying the GGgenotype (180.6 mg/dL vs. 148.3 mg/dL, PG9E = 0.046). The C allele of rs2464196 (HNF1A) interacted with currentsmoking for elevated TG levels (PG9E = 0.028). The geneand cigarette smoking interactions shown in this study needto be validated in a large-scale study.
