The plant cell platform is an efficient system to produce recombinant proteins along with animal and microbial systems. Epidermal growth factor (EGF) is a protein that stimulates cell growth by binding to its receptor. This protein has been used for medicinal purposes such as wound healing. Several studies have shown that the production of recombinant proteins increases when targeting the endoplasmic reticulum in various cell organelles. Herein, we introduced EGF and EGF tailed with KDEL (EGF-KDEL), a signal peptide targeting the endoplasmic reticulum, in Centella asiatica cells. The optimal culture conditions of transgenic C. asiatica callus were investigated in two selected transgenic cell lines in a 3 L air-lift bioreactor. To analyze the efficacy of EGF produced from the transgenic cell culture, human keratinocyte HaCaT cells were treated with the soluble proteins extracted from these cells. After transformation by Agrobacterium, EGF and EGF-KDEL were successfully integrated into the genome of C. asiatica cells, and subsequently, two transgenic cell lines (E3 among EGF cell lines and EK2 among EGF-KDEL cell lines) were selected based on EGF expression level, cell growth, and total soluble protein content.
