Viral haemorrhagic septicaemia (VHS) is serious viral diseases of farmed salmonid, turbot and olive flounder as well as many fish species of freshwater and marine water, so the VHS is listed disease by world organization for animal health (OIE). The causative agent of the disease, VHS virus (VHSV) is classified into four genotypes (I to IV) by phylogenetic analyses from sequence of the nucleocapsid (N) and glycoprotein (G) of the virus, and it was associated with geographical location.
Various diagnostic methods for VHS have been described in the OIE manual of diagnostic test, including infectious virus isolation by cell culture method, viral protein detection by enzyme- linked immunosorbent assay (ELISA), viral gene detection by conventional reverse-transcription (RT)-PCR and real time RT-PCR. Among them, the conventional RT-PCR method has been commonly used for viral gene detection and phylogenetic analysis of sequencing from the PCR product.
