Caspase-8 (Casp8) is one of the important initiator caspases, categorized under the cysteinyl aspartate proteinase family proteins. It triggers the intrinsic and extrinsic apoptosis pathway to eliminate the infected, damaged, or mutated cells from the organisms. In the current study, we have isolated the Casp8 gene from the previously constructed Amphiprion clarkii (AcCasp8) NGS database and characterized using in-silico analysis. Then challenge experiment was performed to distinguish the defensive capability of AcCasp8 against the live bacteria (Vibrio harveyi) and PAMPs (Poly I:C and LPS). Also, down regulatory pathway gene expression was observed after the transfection of AcCasp8 to the HEK293T cells. The complete cDNA sequence of AcCasp8 was 1872 bp and the coding region consisted of 1461 bp, encodes 486 amino acids (aa).
