The AvChit gene encodes for a chitinase from the spider,Araneus ventricosus. This spider, A. ventricosus, isan abundant species in Korea. Arabidopsis thalianaplants were transformed with the AvChit gene usingAgrobacterium tumefaciens. Thirteen transgenic linesexpressing the AvChit gene were obtained. Functionalexpression of the AvChit gene in transgenic Arabidopsiswas confirmed by Southern, northern and westernblot analysis. The AvChit cDNA was expressed as a61 kDa polypeptide in baculovirus-infected insect Sf9cells. AvChit protein extracted from transgenic Arabidopsisexhibited high levels of chitinase activity. Phytopathologicaltests showed that two transgenicArabidopsis lines expressing the AvChit gene displayedhigh levels of resistance to gray mold disease (Botrytiscinerea).
