In radish, self-incompatibility(SI) is governed by the S-locus, which consists of a series of multiple S-alleles. Most of the commercial F1 hybrid seeds are produced using the SI system. The present investigation was performed with 88 inbred lines which were used as parents of commercial F1 hybrids. DNA fragments of SLG alleles were amplified from all inbred lines by PCR using SLG universal primer pair USf and USr. Digestion of the PCR products with restriction endonucleases and subsequent Mataphor agarose gel electrophoresis revealed polymorphism of the amplified DNA fragment. Electrophoretic profiles of the PCR products after digestion with HinfI and MspI were classified into 11 different types. The numbers of S-haplotypes detected in the inbred lines were 11(S1~S11) and the highly frequent S-haplotypes were S7(30.6%), S3(23.8%), and S11(11.3%). Our investigation showed that the PCR-RFLP analysis using universal primer pairs of SLG was useful for identifying the S-haplotypes in radish. To estimate the activity of SI of S-haplotypes identified by seeds set analysis, we calculated the number of seed setting, which corresponds to the ratio between number of seeds obtained and number of siliques after self-pollination. The number of seed setting varied from 0 to 4.3 in all inbred lines. The average number of seed setting showed any significant statistical differences among different lines. The activity of SI for S-haplotype appeared to differ according to the genetic background of inbred lines. Consequently, it was expected that there exist other factors related to the activity of SI.
