The somatic hybrids between Solanum brevidens and S. tuberosum (cv. Superior and Dejima) can be selected through thenumber of chloroplasts in the guard cells, SSR (simple sequence repeats) and AFLP. In order to facilitate the identification of hybrids atearly stage following protoplast fusion, four microsatelite markers were selected with the size of 21, 44, 48, or 51 bps. The size of theamplified DNA fragments ranged from 110 to 480 base pairs. Seven combinations of primers were selected for the AFLP markers. Anaverage of 33.1 bands per AFLP primer were generated in each parent with an average 21.3 of these polymorphic bands. The data ofSR and AFLP analysis confirms the hybrid nature of analyzed plants; the somatic hybrids displayed combination of specific parentalbands. The SSR and AFLP markers provide a quick, simple and preliminary screning method for putative somatic hybrids. Putativesomatic hybrids could also be selected through the number of chloroplasts in the guard cels. which ranged from 16 to 19.
