A sensitive optical waveguide lightmode spectroscopy-based immunosensor was developed to detect vitelogenin inseawater flatfish (Paralichthys olivaceus). For this purpose,anion-exchange column chromatography with DE-52 resinwas used to purify flatfish vitelogenin from flatfish serumcontaining vitellogenin that had ben induced using anintraperitoneal 17β-estradiol injection. The anti-flatfish vitellogeninimmunosensor was prepared using the purified flatfishvitellogenin. The change in the incoupling angle acording tothe complexation between the flatfish vitellogenin and itsantibody, immobilized over an optical grating coupler sensorchip, was measured to calculate the sensor response. Theimmunosensor was quite specific to flatfish vitellogeninbinding, based on no sensor response in the case of bovineserum albumin immobilization. When plotted using double-flatfish vitellogenin concentrations of 0.00675-67.5 nM, witha detection limit of 0.0675 nM. The reusability during sevenrepetitive measurements was reasonably fair for the preliminaryscreening of flatfish vitellogenin.
