Vibrio vulnificus is an opportunistic pathogen thatcauses septicemia in humans. To identify the genes associatedwith its pathogenicity, in vivo expresion technology (IVET)was used to select genes specificaly expressed in a host, yetnot significantly in vitro. Random lacZ-fusions in the genomeof V. vulnificus strain MO6-24/O were constructed using anpromoterles-aph and -lacZ as reporter genes. A total of~18,0 resulting library clones were then intraperitoneallyinjected into BALB/c mice using a colony forming unit(CFU) of 1.6×106. Two hours after infection, kanamycin wasadministered at 20g per gram of mouse weight. After twoselection cycles, 11 genes were eventually isolated, which wereexpressed only in the host. Among these genes, VV20781 andVV21007 exhibiting a homology to a hemagglutinin gene andtolCfrequency. When compared to wild-type cells, mutants withlesions in these genes showed no diference in the rate ofgrowth rate, yet a significant decrease in cytotoxicity and thecapability to form a biofilm.
