Polyhydroxyalkanoates (PHAs) are homo or heteropolyesters of (R)-hydroxyalkanoates accumulated in variousmicroorganisms under growth-limiting condition in the presenceof excess carbon source. They have been suggested asbiodegradable substitutes for chemically synthesized polymers.Recombinant Escherichia coli is one of the promising hoststrains for the economical production of PHAs, and has beenextensively investigated for the process development. Theheterologous PHA biosynthetic pathways have been establishedthrough the metabolic engineering and inherent metabolicpathways of E. coli have been redirected to supply PHAprecursors. Fermentation strategies for cultivating theserecombinant E. coli strains have also been developed for theefficient production of PHAs. Nowadays, short-chain-length(SCL) PHAs are being re-invited due to its improvedmechanical properties and possible applications in thebiomedical area. In this article, recent advances in thedevelopment of metabolically engineered E. coli strains forthe enhanced production of SCL-PHAs are reviewed. Also,medical applications of SCL-PHAs are discussed.
