An ABC transporter apparatus of the Gram-negativebacterial type I secretion pathway can be used as a secretoryprotein expresion system in Escherichia coli. Four types ofcoexpression systems for the Pseudomonas fluorescens lipasegene, tliA, and its cognate ABC transporter gene cluster,tliDEF, were constructed. When the relative expresion levelswere changed by adding diferent concentrations of IPTG, thesecretion (16.9 U/ml of culture) of TliA in E. coli [pTliDEFA-223+pACYC184] was significantly higher than E. coli[pKK223-3+pTliDEFA-184] secreting the lowest level ofTliA (5.2 U/ml of culture). Maximal acumulation of thelipase secreted occurred in the mid-exponential phase, implyingthat the efficient protein secretion via an ABC transporter waslevel of TliA in E. coli [pTliDEFA-223+pACYC184] wasincreased to 26.4 U/ml by inducing gene expresion at theculture initiation time. These results indicate that a significantincrease in the ABC transporter-dependent protein secretioncan be achieved by simply controling the relative expressionlevels between the ABC transporter and its pasenger protein,even in the recombinant E. coli cells.
