UDP-N-Acetylglucosamine(GlcNAc):β1,4-Dmannosideβ-1,4N-acetylglucosaminyltransferase-III (GnTIII)and UDP-N-GlcNAc:α-6-D-mannosideβ-1,6Nacetylglucosaminyltransferase-V (GnT-V) activities weredetermined in human hepatoma cell lines and metastatic coloncancer cells, and their activities were compared with those ofnormal liver cells and fetal hepatocytes. GnT-III activitieswere higher than those of GnT-V in hepatic carcinoma cells.When the two enzyme activities were assayed in highlymetastatic colon cancer cells, GnT-V activities were muchhigher than those of GnT-III. When GlcN,GlcN-biant-PA andUDP-GlcNAc were used as substrates, the enzymes displayeddifferent kinetic properties between hepatic and colon cancercells, depending on their metastatic potentials. Normal cellsof two origins had characteristically very low levels of GnTIIIand -V activities, whereas hepatoma and colon cancer cellscontained high levels of activities. These data were supportedby RT-PCR and Northern blot analyses, showing that theexpression of GnT-III and -V mRNAs were increased inproportion to the enzymatic activities. The increased GnT-IIIand -V activities were also correlated with increased glycosylationof the cellular glycoproteins in hepatoma and colon cancercells, as examined by lectin blotting analysis by using wheatgerm glutinin (WGA), erythroagglutinating phytohemagglutinin(E-PHA), leukoagglutinating phytohemagglutinin (L-PHA), andconcanavalin A (Con A). Treatment with retinoic acid, adifferentiation agent, resulted in decreases of both GnT-IIIand -V activities of HepG2 and HepG3 cells. In colon carcinomacells, however, treatment with retinoic acid resulted in areduction of GnT-V activity, but not with GnT-III activity.Although the mechanism underlying the induction of theseenzymes is unclear, oligosaccharides in many glycoproteinshave been observed of cancer cells.
