Nonsteroidal anti-inflammatory drugs such asindomethacin induce severe gastric mucosal damage in humansand rodents. In the present study, the in vivo protectiveeffect of astaxanthin on indomethacin-induced gastric lesionsin rats was investigated. The test groups were injectedwith indomethacin (25 mg/kg) after the oral administrationof astaxanthin (25 mg/kg) for 1, 2, and 3 days, while thecontrol group was treated only with indomethacin. Thiobarbituricacid reactive substances in the gastric mucosa, as an index oflipid peroxidation, increased significantly after indomethacinadministration and this increase was inhibited by oraladministration of astaxanthin. In addition, pretreatment withastaxanthin resulted in a significant increase of the activitiesof superoxide dismutase (SOD), catalase, and glutathioneperoxidase (GSH-px). Histologic examination clearly revealedacute gastric mucosal lesions induced by indomethacin in thestomach of the control group, but were not observed in that ofthe test group. These results indicate that astaxanthin activatesSOD, catalase, and GSH-px, and removes the lipid peroxidesand free radicals induced by indomethacin. It is evident thatastaxanthin acts as a free radical quencher and antioxidant,and is an effective molecule in the remedy of gastric mucosallesions.
