Fucoidan is a sulfated polysaccharide found in algae, including Fucus vesiculosus. Several studies have demonstrated the biological effects of fucoidan, which include anti-viral activity and the amelioration of allergic diseases. In addition, it has been shown that fucoidan stimulates dendritic cells and protects bone marrow cells from ionizing radiation. However, the effects of fucoidan on splenocytes, which play a critical role in the immune system, have not yet been elucidated. In this study, a proliferation assay showed that fucoidan stimulated splenocyte proliferation, while flow cytometric analysis demonstrated that fucoidan increased the size of splenocytes, as compared to control cells. To further analyze the changes in the fucoidan-treated splenocytes, the expression levels of the lymphocyte surface markers CD4, CD8, and CD19 were measured. There was no significant change in lymphocyte surface marker expression between the control and fucoidan-treated splenocytes. Propidium iodide staining of the cells revealed that fucoidan increased splenocyte viability. Furthermore, the effect of fucoidan on the clonal expansion of splenic lymphocytes was investigated. The fucoidan-treated splenocytes showed increased expression of CD25, the alpha chain of interleukin (IL)-2 receptor, which is a marker of lymphocyte activation; moreover, they responded strongly to IL-2, a major cytokine in clonal lymphocyte expansion. Taken together, these findings demonstrate that fucoidan has immunostimulatory effects on splenocytes.
