Sodium selenite (selenium) is a dietary essential trace element. Selenium has roles as an antioxidant and redox regulator. We determined whether the prophylactic injection of selenium would protect the intestinal mucosa from irradiation injury in rats. Selenium (0.5 ㎎/㎏) was administered via an intraperitoneal injection every 48 h for 7 days prior to irradiation. The abdomen was exposed to gammaradiation with a single dose of 12 Gy. The entire small intestine from the duodenum to the ileum underwent histologic evaluation. We selected the ileum for semi-quantitative evaluation because lesions of the ileum had more severe injuries compared to the proximal portion. We performed the TUNEL stain on the 1<SUP>st</SUP> day and evaluated the mucosal injury using a semi-quantitative scoring system on the 5<SUP>th</SUP> day. Apoptotic cells in the untreated group were frequently detected compared to the selenium-treated group (22±1/50 crypts vs. 7.8±1.2/50 crypts, P<0.05). The mucosal injuries were severe in the untreated group, whereas the selenium-treated group significantly decreased the severity of the injury. The damaged area in the untreated group was wider than the selenium-treated group. The histopathologic examination revealed that the intestinal mucosa was preserved, and the number of apoptotic cells was decreased in the selenium-treated group compared to the untreated group. The current study showed that the ileum is the most sensitive organ among the small intestine and administering selenium decreased radiationinduced mucosal injuries in the intestine after irradiation. In conclusion, prophylactic administration of selenium might reduce irradiation-induced ileal mucosal damage.
