There are 35 kinds of herpes virus in macaques, and among them, only B virus is known to have pathogenicity in human among them. Although B virus infection is generally asymptomatic or mild in macaques, it can cause fatal encephalomyelitis in human. Herpes simplex virus-1 (HSV-1) has been used as alternative antigen to replace dangerous B virus because of its high genetic similarity. However, the method has limitation because of its high false positive reaction to other Herpes viruses including Herpes simplex virus-2 (HSV-2). Therefore, more specific and safer antigen is urgently needed to replace HSV-1. In this study, we performed Simian Agent 8 (SA-8) antigen-based enzyme-linked immunosorbent assay (ELISA) to confirm if this antigen has a higher specificity to B virus compared to HSV-1. We collected sera from 136 Cynomolgus monkeys on the day of acceptance and 30th day after acceptance and these samples were tested using HSV-1 and SA-8 based ELISA. In the HSV-1 based ELISA test, 10 and 7 samples were positive or equivocal on day 0 and day 30 respectively. We tested the positive or equivocal sample of day 30 using SA-8 antigen based ELISA test. HSV-1 positive or equivocal samples were tested at The Corporation for Production and Research of Laboratory Primates in Tsukuba, Japan which established the B virus antigen based ELISA. And they all turned out to be negative. The results from this study show that SA-8 virus has higher specificity against B virus compared to HSV-1 and can be used as an alternative antigen to replace it.
