The objective of current study is to examine the effects of conjugated linoleic acid (CLA) on the change of peripheral blood cells, the phagocytic response of peripheral blood leukocytes and the plasma tumor necrosis factor (TNF)-α level in rats fed a diet with CLAs. Male 10-week-old Sprague-Dawley rats were fed on powder diets containing 0.75% either CLA mixture (45% cis-9, trans-11 (9c,11t)-CLA and 43% trans-10, cis-12 (10t,12c)-CLA) or 10t,12c-CLA for 8 days. The change of blood cell values (RBC, PCV, WBC, differential count of WBC and platelets), the phagocytic activity of WBC, and the plasma TNF-α level were evaluated on days 0, 4, 8 and 12, respectively. There were no significant changes in the numbers of RBC, platelets and the value of PCV regardless of CLA supplement. But, the numbers of WBC and neutrophils on the differential count of WBC in rats fed a diet with either CLA mixture or 10t,12c-CLA were significantly increased (p<0.01) on day 8 when compared with those of control rats fed a diet without CLAs. The phagocytic activity of WBC was remarkably increased on day 4 (p<0.05) and peaked on day 8 (p<0.01). Thereafter the activity trended to decrease on day 12 (p<0.01) but it was still very high when compared with that of control rats. This similar finding also showed in peripheral blood polymorphonuclear cell (PMN)-rich cells fractioned from flowcytometric cytography. Plasma TNF-α level measured by TNF-α sensitive cell line WEHI-164 bioassay was increased on day 4 and it showed a significant increase (p<0.01) on day 8 when compared with that of control rats. These results suggested that the supplement of CLAs to rats induces an enhancement of PMN phagocytosis, which may be associated with the increase of plasma TNF-α.
