High level of blood cholesterol is one of the most important risk factor related with coronary heart disease and atherosclerosis. The main objective of this study was to investigate the cholesterol-lowering effect of lactic acid bacteria (LAB) isolated from human intestine and rutin extracted from buckwheat in high cholesterol-diet fed Sprague-Dawley rats. In addition, it was determined whether intestinal microflora composition is improved by LAB and/or rutin consumption. Five-week old male rats were administrated with 0.59 ㎎/㎏ B.W. of rutin and/or 1.5 × 10?-6 × 10? cfu/㎏ B.W. of LAB as an oral route with cholesterol diet for 6 weeks. Total cholesterol, triglyceride, low-density lipoprotein (LDL)-cholesterol, and high-density lipoprotein (HDL)-cholesterol levels in blood were determined biweekly. The levels of total cholesterol, triglyceride and LDL-cholesterol were significantly decreased in rats administrated with rutin (Group A) compared to Positive control group at 4 weeks. Rats administrated with rutin and LAB (Group C) showed also a significantly reduced triglyceride and the ratio of LDL against total cholesterol, and increased the ratio of HDL. The relative liver weights were significantly decreased in all treated groups compared to positive control group at 6 weeks. In bacteriological analysis, the number of Streptococcus and Lactobacillus was significantly increased in rats administrated with LAB (Group B). On the other hand, the number and the frequency of occurrence of Enterobacteriaceae were significantly decreased in Group B. This study suggested that combined administration of rutin and LAB has a hypocholesterolemic effect and could be used to prevent coronary heart disease and atherosclerosis as well as fatty liver. In addition, they would make an improvement of the intestinal microflora composition by decreasing putrefactive bacteria and increasing LAB.
