Single-dose toxicity of DA-3050, a recombinant human basic fibroblast growth factor (bFGF), was studied in SD rats via oral, subcutaneous and intravenous routes and in beagle dogs via subcutaneous routes. In addition, acute subcutaneous toxicity of heat-degraded bFGF in rats was investigated. In all experiments, no mortality was observed. In rats received bFGF intravenously, significant decrease in body weight gain and soft stools were observed. Swelling of injection site was the main clinical sign in subcutaneously treated rats. In dogs, reductions in body weight, locomotion and food consumption were observed in bFGF injected animals. White blood cell count slightly decreased and platelet count markedly decreased in 5 mg/kg group. In the serum biochemical analysis, inorganic phosphorus, creatinine and blood urea nitrogen decreased and cholesterol and triglyceride increased in 5 mg/kg dosing group. At necropsy, subcutaneous swelling, congestion, edema and focal hemorrhage were noted at the injection site. Histological examination of the injection site showed infiltration of fibroblasts and inflammatory cells, and diffuse neovascularization, which were more predominant in 5 mg/kg groups. One male dog in 5 mg/kg group showed focal interstitial nephritis and glomerular epithelium vacuolation in the kidneys. Based on the results, the LD50 values of bFGF were estimated to be greater than 20 mg/kg (p.o. and s.c.) or 10 mg/kg (Lv.) in rats and greater than 5 mg/kg (s.c.) in dogs under the current test conditions. The minimum lethal dose of heat-degraded bFGF was also greater than 20 mg/kg (s.c.) in rats.
