Studies were carried out to compare the toxic potency of 3,3',4,4',5-pentachlorobiphenyl (PCB 126) with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) in newly fertilized Japanese medaka (Oryzias latiper) eggs and one-month-old larvae using several toxicological end points based on the tissue doses. PCB 126 caused the same type and sequence of lesions as TCDD in the embryos and newly hatched larvae. The toxic effects in the cardiovascular system were the first to appear, and the typical lesions observed were
multifocal hemorrhages, pericardial and yolk sac edema, craniofacial malformation, and inhibition of swim bladder inflation (SBl). The inhibition of SBI was the most sensitive and clearly defined end point measured in the embryo-larval assay, and the ED50 (pg/mg embryo) values for TCDD and PCB 126 were 1.9 and 59.2, respectively. The wasting-type syndrome was the most sensitive end point measured in the one-month-old larval assay, and the ED50 (pg/mg larva) values for TCDD and PCB 126 were 2.0 and 89.7, respectively. The toxic equivalency factor (TEF) for PCB 126 based on LD50 values ranged from 0.017 to 0.024, and these values are much different from those reported for trout species and in the range reported for mammalian species. These TEF values might be useful in assessing the risk of TCDD and PCB 126 to fish early life stage survival.
