The purpose of this study was to investigate the effect of Lonicera Japonica Thunberg(LJT) on inhibiton of inflammatory factor expression associated with atherosclerosis in HUVEC.
At first, we measured safety in cytotoxicity of LJT. After treatment with LJT in HUVEC, we measured the expression levels of MCP-1, ICAM-1, VCAM-1, KLF2, eNOS, ERK, JNK, p38 proteins, and the expression level of CCL2, ICAM1, VCAM1, KLF2, NOS3 mRNA.
Results were summarized as follows :
1. LJT did not show any cytotoxicity in HUVEC within the concentration tested except for a concentration of 600㎍/㎖.
2. LJT significantly reduced MCP-1, VCAM-1 levels in HUVEC at concentrations of 100, 200, 400㎍/㎖ compared to control group, and significantly reduced ICAM-1 level in HUVEC at concentrations of 200, 400 ㎍/㎖ compared to control group. Also LJT increased KLF2 level in HUVEC at concentrations of 100, 200, 400㎍/㎖ compared to control group but it was not significant. And LJT significantly increased eNOS level in HUVEC at concentration of 400㎍/㎖ compared to control group.
3. LJT significantly reduced CCL2, ICAM1, VCAM1 mRNA expression in HUVEC at concentrations of 100, 200, 400㎍/㎖ compared to control group, and significantly increased KLF2 mRNA expression in HUVEC at concentration of 400㎍/㎖ compared to control group, and significantly increased NOS3 mRNA expression in HUVEC at concentrations of 200, 400㎍/㎖ compared to control group,
4. LJT significantly reduced MCP-1, VCAM-1 protein expression in HUVEC at concentrations of 200, 400㎍/㎖ compared to control group, and significantly reduced ICAM-1 protein expression in HUVEC at concentration of 400㎍/㎖ compared to control group. Also LJT increased KLF2, eNOS protein expression in HUVEC at concentrations of 200, 400㎍/㎖ compared to control group. LJT did not have an effect on ERK protein expression compared to control group. LJT significantly reduced JNK protein expression in HUVEC at concentrations of 200, 400㎍/㎖ compared to control group, and significantly reduced p38 protein expression in HUVEC at concentration of 400㎍/㎖ compared to control group,
These results suggest that LJT has an effect on inhibition of inflammatory factor expression associated with atherosclerosis in HUVEC, which helps the prevention of cardiovascular and cerebrovascular diseases.
