Objectives : This study is aimed to evaluate the protective effect of Gastrodiae Rhizoma and steamed, dried & fermented Gastrodiae Rhizoma on LPS-induced hepatic injury in the mice model.
Methods : Mice were randomly divided into 4 groups-Normal group, vehicle group (LPS treated), GR0F0 group (fed GR0F0 - not processed Gastrodiae Rhizoma, before LPS treating) and GR6F4 group (fed GR6F4-fermented with #4 strain and Gastrodiae Rhizoma that steamed and dried 6 times, before LPS treating) with 6 mice in each group.
GR0F0 group and GR6F4 group were fed each material 200 ㎎/㎏/day during 8 days. Then they maintained NPO state for 24 hours. LPS 20 ㎎/㎏ injected to the experimental groups as abdominal injection.
Blood samples were collected 24 hours after LPS injection for liver function and enzyme activity and the livers were harvested for histo- logical, immuno-histochemical and western blot analysis.
Results : The activity was high in order of GR6F4 and GR9F0 (non-fermented Gastrodiae Rhizoma that steamed and dried 9 times) in DPPH radical scavenging activity measurement. In ABTS radical scavenging activity, GR0F0, GR6F4 were a higher order of activity.
Gastrodin was measured a lot in steamed & dried Gastrodiae Rhizoma, gastrodigenin was determined a lot in fermented Gastrodiae Rhizoma. Samples containing the most effective component was GR6F4.
Toxicity was not found in GR0F0 and GR6F4.
Measurement of serum ALT and AST in laboratory animals to induce liver damage by LPS, GR0F0 and GR6F4 showed a significant decrease compared to the vehicle group. As a result of measuring the ROS, GR6F4 showed a significant reduction in both the serum and liver tissues compared to the vehicle group. GR0F0 group showed a significant reduction only in the liver tissues.
AP-1, COX-2, iNOS were significantly decreased GR0F0 group and GR6F4 group. But TNF-α only showed a significant reduction in GR6F4.
Conclusion :
GR0F0, GR6F4 against liver demage in mice with LPS showed significant effects on anti-oxidant and anti-inflammatory action. The effects of GR6F4 showed superior results compared to GR0F0. Therefore, Gastrodiae Rhizoma and steamed, dried & fermented Gastrodiae Rhizoma were thought to have a protective effect on liver injury.
