In this study, we introduced polyamidoamine (PAMAM) generation 4 conjugated with cell penetrating peptides (CPPs) consisting of Lysine-Phenylalanine-Phenylalanine (G4-KFF, G4-FFK) amino acid sequence to develop an outstanding nano-carrier for gene delivery. Cell penetrating peptides are well known to deliver biomolecules effectively into the cell, and believed to be originated from PTD (Protein-Transduction Domain) or MTS (Membrane-Trans locating Sequence) from virus. Generally, it has special cellular uptake mechanism without any damage to the cell membrane. For this reason, we conjugated cell penetrating peptides on the surface of PAMAM dendrimers in order to advantage of this property. DNA condensation efficiency of G4-KFF and G4-FFK was confirmed by agarose gel retardation assay. Then size of prepared polyplexes was analyzed by DLS and observed STEM and TEM image. Then cytotoxicity was evaluated in different cell lines; HeLa and HepG2 cells. We found transfection efficiency was remarkably higher in G4-KFF and G4-FFK in both the cell line compared with gold standard PEI 25 kDa transfection reagent with negligible cytotoxicity. Here we believe that our nano-carrier will be a promising non-viral vector to deliver therapeutic genes due to its greater cellular uptake mechanism.
