This study is aimed at investigating the protective effect of Ganoderma lucidum pharmacopuncture (GLP) on hepatotoxicity induced acutely and chronically by ethanol (EtOH) in rats.
SD Rats were divided into 4 groups of 8 animals each: normal, control, NP (Normal saline pharmacopuncture) and GLP. Control, NP and GLP groups received ethanol (acute & chronic). NP and GLP groups were daily treated with injection of normal saline and Ganoderma lucidum extract respectively. Control group received no treatment.
As a result, GLP reduced the histological changes induced by EtOH in both acute and chronic liver injury. GLP significantly reduced activities of ALT enzymes increase induced by EtOH for 6h. In test for 4 weeks of EtOH administration, both AST and ALT enzymes of GLP group were significantly reduced. GLP significantly ameliorated SOD and CAT activity in both acute and chronic injury. And GLP significantly decreased BAX and increased Bcl-2 immunoreactivity expression in chronic liver injury.
In conclusion, the present study indicated the protective efficacy of GLP treatment against EtOH-induced hepatic injury in SD rats by modulating ethanol metabolizing enzymes activity, attenuating oxidative stress, and inhibiting mitochondrial damage-mediated apoptosis.
