Ascorbate peroxidase (APX), a gene, protects plants from abiotic stress by regulating reactive oxygen species levels. Although its function has been studied in model plants, research on APX in zoysiagrass is limited. This study identified nine APX1 and five APX2 genes in four zoysiagrass species (Zoysia japonica, Z. pacifica, Z. sinica, and Z. matrella) using bioinformatic tools. Regulatory region analysis confirmed that APX1 has a transcription start site (TSS) with a TATA box, whereas APX2 mostly has a TATA-less TSS. Transcription factors (TFs) related to stress regulation, especially ERF, appeared most frequently; APX2 genes were predicted to have an abundance of TFs, indicating a greater dependence on regulatory elements for stress responses. Protein structure prediction analysis revealed changes in the protein structure due to insertions and single amino acid polymorphisms; phylogenetic analysis supported these findings. In particular, the predicted deficiency in the heme-binding structure of APX1 in Z. japonica and Z. matrella suggests differences in their stress responses. This study provides a genetic foundation to enhance our understanding of the environmental adaptation mechanisms and stress resistance of the four zoysiagrass species.
