Matricellular proteins are integral non-structural components ofthe extracellular matrix. They serve as essential modulators ofimmunometabolism and tissue homeostasis, playing critical rolesin physiological and pathological conditions. These extracellularmatrix proteins including thrombospondins, osteopontin,tenascins, the secreted protein acidic and rich in cysteine (SPARC)family, the Cyr61, CTGF, NOV (CCN) family, and fibulins havemulti-faceted functions in regulating immune cell functions,metabolic pathways, and tissue homeostasis. They are involvedin immune-metabolic regulation and influence processes suchas insulin signaling, adipogenesis, lipid metabolism, and immunecell function, playing significant roles in metabolic disorderssuch as obesity and diabetes. Furthermore, their modulationof tissue homeostasis processes including cellular adhesion,differentiation, migration, repair, and regeneration is instrumentalfor maintaining tissue integrity and function. The importanceof these proteins in maintaining physiological equilibriumis underscored by the fact that alterations in their expressionor function often coincide with disease manifestation. Thisreview contributes to our growing understanding of these proteins,their mechanisms, and their potential therapeutic applications.
