Minoxidil and Finasteride are the only drugs approved by the FDA and used as treatments for alopecia, but interest in natural products with similar effects and less side effects is increasing as treatment using them is incomplete and questions about safety and potential side effects are raised. In this study, three kinds of plant extracts(Serenoa repens, Aloe saponaria, Aloe vera) were examined for antioxidant activity and, human and rat dermal papilla cells proliferation. Aomng them, Aloe saponaria was chosen as the best candidate material. For in-depth study on Aloe saponaria, which has not yet been studied, antioxidant tests, rat and dermal papilla cell proliferation, and protein expression analysis were conducted using solvent fractions. In human dermal papilla cells, Aloe saponaria extract increased cell proliferation by 28% at 25 ㎍/㎖, and in rat dermal papilla cells, it increased cell proliferation by 36% and 40% at 25 ㎍/㎖ and 50 ㎍/㎖, respectively. In human dermal papilla cells, Aloe saponaria chloroform and ethyl acetate fractions increased cell proliferation by 30% and 8% in 5 ㎍/㎖, respectively. In rat dermal papilla cells, Aloe saponaria chloroform and ethyl acetate fractions increased cell proliferation by 53% and 32% in 25 ㎍/㎖, respectively. There was no significant increase in dermal papilla cells in butanol fraction and water fraction. As a result of protein expression analysis, expression and phosphorylation of AKT were not detected, but phosphorylation of ERK was increased in Aloe saponaria extract, Aloe saponaria chloroform fraction, and Aloe saponaria ethyl acetate fraction, thereby promoting the growth of dermal papilla cells. Since there is no prior research on Aloe saponaria, it is reasonable to believe that it can be developed into a new natural substance that is effective in hair growth, hair loss prevention and treatment.
