This study was conducted to establish adventitious roots (AR) culture from the original seeds of wild plants Allochrusa gypsophiloides (Regel) Schischk, a Central Asian rare species, whose roots are a rich source of oleanane-type triterpene saponins. The root explants from in vitro raised seedlings were cultured on liquid Murashige and Skoog medium (MS) with 1 mg/L indole-3-butyric acid (IBA) or α-naphthalene acetic acid (NAA) under continuous agitation and total dark condition during a 45-day cycle. The AR induction, fresh (dry) root weight, growth index, and total saponins content in culture were evaluated. The high proliferative activity of AR on an auxin-containing medium resulted in accumulation and increased growth of AR relative to control, with a significant 3.5-fold excess on ½ MS with IBA. An increase in biomass AR on IBA and NAA medium correlated with the increased saponins content in their ethanol extracts compared to hormone-free. The excess of saponins in vitro on auxin-containing medium relative to in vivo roots averaged 138%, on control medium the saponins content was 1.5 timers lower. For the first time, the AR culture in vitro with a high growth index and high produc- tion of saponins for A. gypsophiloides was obtained.
