Sphaerodes mycoparasitica is a mycoparasite that attacks major plant pathogenic Fusarium species including F. avenaceum (Fr.) Sacc., F. oxysporum Schltdl., and F. graminearum Schwabe. Especially, F. graminearum complex is a devastating causal agent of Fusarium Head Blight (FHB) in cereal crops such as wheat and barley. FHB has continued significant losses in seed germination, grain yield, and grain quality resulting in negative impacts on global food safety and security, which merits scientific attention and carrying out research. With the growing need for an environmentally-friendly solution (biological control), in our research, S. mycoparasitica as a potential biocontrol agent was investigated for the ability to control plant pathogenic and mycotoxigenic Fusarium species.
In the initial stage of this research, the mycoparasite-host cell-to-cell replacement and interface changes in F. graminearum, as well as a decrease in the production of mycotoxins in co-culture systems, gave a hint of a very complex fungus-fungus interrelationship. Recent studies demonstrated that the level of S. mycoparasitica adaptability strongly depends on types of Fusarium filtrates indicating the degree of host compatibility ranging from biotrophic-attraction to antagonistic-inhibition relationships. Further, the capacity of the mycoparasite to change the hydrophobicity of the host fungal surfaces during mycoparasitism was supportive of this finding. As an important feature, S. mycoparasitica possessed the ability to degrade or transform Fusarium mycotoxins such as zearalenone, 15-acetyl-deoxynivalenol, 3-acetyl-deoxynivalenol, and deoxynivalenol.
On top of that, research on S. mycoparasitica through the tripartite system, S. mycoparasitica (biocontrol agent)-F. graminearum (plant pathogen)–Triticum durum (plant host) provided new insight into the interactome. In vitro wheat germination assay showed the improved seed germination rate (%) when the seed was protected by S. mycoparasitica against virulent F. graminearum which implied that an early preventive biocontrol measure is possible to suppress the establishment of FHB pathogens in cereal crops. During the mycoparasitism, the interactive hyphae were in action captured by Scanning Electron Microscope and Confocal Laser Scanning Microscopy. The biochemical changes in the roots of the seed germinants during the tripartite interaction were demonstrated by Fourier-Transform Infrared Spectroscopy. Moreover, interactive transcriptomics based on Illumina HiSeq contributed to a better understanding of the mycoparasite and its lifestyle. It was found that gene expression of a filamentous hemagglutinin/adhesion, glucanase, chitinase, and ATP-Binding Cassette transporter was upregulated during the mycoparasitism; whose function is related to the attachment, the penetration, and the degradation of mycotoxins, respectively.
Overall, S. mycoparasitica could be considered as a potential BCA specifically to reduce Fusarium-associated risks. Further proteomics and metabolomics based on transcriptomics would deepen the knowledge on this specific mycoparasite and its mycoparasitism.
