Psoriasis is a chronic inflammatory skin disease and immune-mediated disease. Several psoriasis animal models have been developed. Sphingolipids have been known to regulate the biological functions and immune cells in skin. In this study, ceramide analysis by LC-MS/MS and inhibition of SphK1 by PF543 in IMQ-induced psoriasis mouse model were investigated.
8-week-old BALB/c mice were housed. The mice were divided into four groups: Vaseline treated control group, imiquimod-treated psoriasis group (IMQ) (3.125 mg/mouse), IMQ plus PF-543 group (1 mg/kg), IMQ plus ethanol group. On the 10<SUP>th</SUP> day, mice were sacrificed to collect plasma, spleen, and skins samples.
The ceramides from plasma and back skin samples were extracted by chloroform: methanol (2:1) mixture. The chloroform phase was dried at a vacuum evaporator and the dry residue was reconstituted in 100 μl MeOH before injecting into LC-MS/MS.
