The pks island, one of the well-known pathogenicity islands (PAI), produces a genotoxin, colibactin, and has garnered much interest in recent studies due to strong correlations between pks island and intestinal diseases. Nonetheless, a low-cost, point-of-care (POC) diagnosis of the pks island has remained challenging. Here, we propose a versatile pks island detection platform using de-novo designed riboregulators, toehold switches, which can be designed to sense arbitrary RNA inputs with wide dynamic range, low crosstalk and high programmability.
