Fallopia sachalinensis (F.Schmidt) RonseDecr. is an herbaceous perennial herb and their rhizomes and leaves are used as medicines for constipation and rheumatism. This study was carried out to identify the seed dormancy type of F. sachalinensis and to explore the conditions for the dormancy break. The seeds were harvested in Ulleungdo, October 2020, then they were dry stored at room temperature after cleaning. The major and minor axis of the seed were 3.18 ± 0.087 and 2.22 ± 0.070 mm, and the weight of one hundred seeds was 2.01 ± 0.035 g. The vigor of the seeds was 66% and had E:S ratio of 0.90 ± 0.015 confirming that the embryos were morphologically mature. The moisture content of the seeds was 17.34 ± 1.018%, and the maximum moisture content 54.74 ± 0.740% was reached on the 3 day of water absorption. Initial germination experiments were performed for 30 days in chambers with different temperatures (15, 20, 25, 30°C) and 25/15°C (16/8h light/dark). Germination was recorded every day except for those in dark. To confirm the germination enhancement through after-ripening and gibberellin, seeds that had been matured at room temperature for 3 months were immersed in distilled water and 500 ppm GA₃ and GA<SUB>4+7</SUB> solution for 3 days each then incubated at 25/15°C chamber. Alternating temperature stratification was performed at autumn temperature (AT, 15/10°C, 16/8h) for 4 weeks, then continued to winter temperature (WT, 4°C, 24h) for 0, 4, 8, 12 weeks and seeds were incubated at 25/15°C chamber.
