Background & Aims: Non-alcoholic fatty liver disease (NAFLD) is an increasing risk factor for hepatocellular carcinoma (HCC). The mechanism of progression of NAFLD is not yet fully understood, hampering the development of effective treatments for NAFLD and NAFLD-associated HCC. Recently, it has been reported that the activity of neurofibromatosis type 2 (NF2), a tumor suppressor and upstream regulator of Hippo kinases, is controlled by epithelial splicing regulatory protein-2 (ESRP2) in hepatocytes. In both NAFLD and AFLD, ESRP2 is suppressed by pro-inflammatory cytokines, thus inactivating NF2 by skipping a specific exon that is critical for NF2 activity and function. This, in turn, promotes dephosphorylation and nuclear localization of YAP, a transcription co-activator of genes involved in cell proliferation and anti-apoptosis.
