Neuropsychiatric disorders such as depression are not yet fully understood for pathological mechanism and there are currently no specific methods to accurately test the severity. While depression affects both women and men, females are more prone to the incidence of depression. Despite the strong sex-associated differences, the majority of animal studies has been performed on male models. Here, we explored the sex-specific depression using a sub-chronic variable stress (SCVS) model of mice, which was observed to induce depression-like behavior in female. Non-targeted metabolomics approach was applied using liquid chromatography and gas chromatography both coupled to mass spectrometric detection, while analytical conditions including the extraction and derivatization conditions were optimized for plasma and whole brain.
