We developed a new label-free and ultrasensitive fluorometric method to detect ribonuclease H (RNase H) based on light-up aptamer. The RNA/DNA chimeric hairpin (HP) promoter is composed of a hairpin structure which has RNA/DNA duplex in its stem. Therefore, target RNase H can recognize and hydrolyze RNA strand in the chimeric HP promoter, thereby releasing T7 promoter sequence which is originally blocked by RNA region in the chimeric HP promoter stem.
