Introduction: Postoperative cognitive dysfunction (POCD) following surgery and anesthesia is a common and severe complication, especially in elderly patients. Also, preexisting cognitive impairment may impart a susceptibility to further cognitive dysfunction not yet been completely elucidated. Thus, we hypothesized that the specific impact of anesthesia on individuals with preclinical Alzheimer’s disease (AD) may make individuals more susceptible to increase the risk of progression of cognitive impairment. The aim of this study was to compare the cognitive impairment between adult and preclinical AD mice model after propofol anesthesia and surgery.
Methods: We conducted abdominal surgery in cognitively presymptomatic (5-month-old) sporadic AD (ApoE4-KI) and age-matched control (C57BL/6J) male mice under propofol anesthesia (170mg/kg) via retro-orbital injections during two hours. Morris water (MWM) and Y maze testing was conducted 2day before and 2, 4, 7 days after surgery. Mean escape latencies and zone alteration percentage were recorded as a major outcome.
