Arabidopsis thaliana tetratricopeptide domain-containing thioredoxin (AtTDX), a heat-stable and plant-specific thioredoxin (Trx)-like protein, is a multi-functional enzyme present in Arabidopsis. The enzyme has been shown to act as a disulfide reductase and chaperone, and the activities were dependent on its oligomeric status. To identify the physiological roles of AtTDX in plant cells, defining its subcellular localization is important. In this study, analysis of AtTDX protein using green fluorescent protein fusion constructs revealed the protein is located in both the cytosol and the nucleus of Arabidopsis. In addition to Western blot analysis, AtTDX protein exhibits predominantly low molecular weight (LMW) form in cytosol, whereas predominates in the high molecular weight (HMW) form in nucleus. AtTDX should play important roles in both nucleus and cytosol to protect against various environmental stresses.
