Background: Although vitamin D analogs have been used inthe topical treatment of psoriasis, their mechanisms of actionare not well understand. Calcitriol, the hormonally activevitamin D3 metabolite, has been demonstrated to exertimmunomodulatory effects in the skin by down-regulatingthe expression of Toll-like receptors (TLRs) and proinflammatorycytokines. Objective: We investigated the effects ofcalcitriol on the expression of TLR2, TLR4, antimicrobialpeptide LL-37, and proinflammatory cytokines in culturedhuman keratinocytes. Methods: The mRNA expressionlevels of TLR2, TLR4, tumor necrosis factor α (TNF-α),interleukin (IL)-1β and LL-37 in cultured human keratinocyteswere measured by real-time polymerase chain reaction(PCR) and reverse transcription (RT). Furthermore, wemeasured supernatant TNF-α levels by an enzyme- linkedimmunosorbent assay (ELISA) to confirm the effects of calcitriolon TLR2 and TLR4. Results: As measured by RT-PCRand real-time PCR, calcitriol was found to suppress thelipopolysaccharide- and ultraviolet B radiation-mediatedinduction of expression of TLRs, LL-37 and proinflammatorycytokines such as TNF-α and IL-1β in normal humankeratinocytes. The supernatant TNF-α levels measured byELISA were also suppressed after treatment with calcitriol. Conclusion: Calcitriol may down- regulate inflammatorystated over-expression of LL-37 and proinflammatory cytokines.
