Escherichia coli has a PhoR-PhoB two-componentregulatory system to detect and respond to the changes ofenvironmental phosphate concentration. For the E. coli W3110strain growing under phosphate-limiting condition, the changesof global gene expresion levels were investigated by usinggenes that are involved in phosphate metabolism wereincreased as phosphate became limited, whereas those of thegenes involved in ribosomal protein or amino acid metabolismwere decreased, owing to the stationary phase response. Theupregulated genes could be divided into temporarily andpermanently inducible genes by phosphate starvation. Atthe peak point showing the highest expression levels ofthe phoB and phoR genes under phosphate-limiting condition,the phoB- and/or phowere investigated in detail by comparing the gene expressionlevels among the wild-type and phoB and/or phoR mutantstrains. Overall, the phoB mutation was epistatic over thephoR mutation. It was found that PhoBR and PhoB wereresponsible for the upregulation of the phosphonate orglycerol phosphate metabolism and high-afinity phosphatetransport system, respectively. These results show the complexregulation by the PhoR-PhoB two-component regulatoryE. coli.
