A protein array was fabricated for a miniaturizedimmunoassay using microcontact printing (CP). Apolydimethylsiloxane (PDMS) stamp with a 5m×5 mdimension was molded from a silicon master developed byphotolithography. Under optimal fabrication conditions, includingthe baking, incubation, and exposure time, a silicon masterwas successfully fabricated with a definite aspect ratio. Anantibody fragment was utilized as the ink for the CP, andtransfered to an Au substrate because of the Au-thiol (-SH)interaction. The immobilization and antibody-antigen interactionserum albumin (HSA) was applied to the protein arrayfabricated with an antibody against HSA, the detection limitwas 100 pg/ml of HSA when using a secondary antibodylabeled with a fluorescence tag. The fabricated protein arraymaintained its activity for 14 days.
