Two kinds of Haematococcus pluvialis cells (greenvegetative cells cultivated under optimal cell culture conditionsand red cyst cells maintained under high light stress conditionsto induce astaxanthin production) were used to investigate theprotein expression profiles by two-dimensional electrophoresis,image analysis, and peptide mass fingerprinting. The cellularaccumulation of astaxanthin was evident after exposure tohigh light intensity and reached the maximum celular level22 proteins were upregulated over 2-fold in the red cyst cellswhen compared with the green vegetative cells and selected forfurther analysis by chemically assisted fragmentation (CAF)-MALDI-TOF sequencing to identify the protein functions.Among 22 diferent spots, several key enzymes specific tothe carotenoid pathway, including isopentenyl pyrophosphateisomerase (IPP) and lycopene β-cyclase, appeared in H.pluvialisand lycopene β-cyclase would appear to be involved withcarotenoid accumulation in the cytoplasm, as these peptideswere preferentially upregulated by high light intensity precedingan increase in carotenoid, and only these forms were detectedin the red cyst cels.
