A gene responsible for the chlorothalonilbiotransformationwas cloned from the chromosomal DNA ofOchrobactrum anthropi SH35B, an isolated bacterium strainfrom soil. We determined the nucleotide sequences and foundan open reading frame for glutathione S-transferase (GST). Thedrug-hypersensitive Escherichia coli KAM3 cells transformedwith a plasmid carrying the GST gene can grow in the presenceof chlorothalonil. The GST of O. anthropi SH35B wasexpressed in E. coli and purified by affinity chromatography.The fungicide chlorothalonil was rapidly transformed by thepurified GST in the presence of glutathione. No significantdifference in the chlorothalonil-biotransformation effect wasobserved among the thiol compounds (cysteine, reducedglutathione, and β-mercaptoethanol). Thus, the result reportedhere is the first evidence on the chlorothalonil-biotransformationby conjugation with the cellular free thiol groups, especiallyglutathione, catalyzed by the bacterial GST.
