Laboratory-based molecular biology instruction modules for high school students were developed to improve their understanding for the concepts related to DNA and genes. Six modules, which can be applied either successively or individually, consist of molecular biology experiments such as DNA extraction from epithelial cells of human oral cavity, agarose gel electrophoresis, amplification of ALDH2 DNA fragment by polymerase chain reaction, restriction enzyme digestion, and observation of single nucleotide polymorphism of ALDH2. To test their applicability and effect for high school students, we applied those to 75 students of 12^th grade in Biology Ⅱ classes and 24 students of 10^th and 11^th grade in ‘after-school classes’, and analyzed their responses through questionnaires. Majority of students responded that these instruction modules were easy to understand and improved their understanding on the relationship among DNA, genes, and proteins. Their attention and interest on molecular biology news on TV, world wide web, or newspapers were also increased. For 10th and 11th graders, these instruction modules influenced students’ mind for future majors and careers. These modules can be reconstructed based on the level of students or laboratory environment. Taken together, the instruction modules would be applicable and useful for several regular or special classes for high school students.
