Background and objectives: Inducible nitric oxide synthase (iNOS) is expressed in some pathological states of the brain, such as cerebral ischemia and traumatic brain injury, and involved in delayed, secondary neuronal injury. However, its involvement in intracerebral hemorrhage (ICH) was unknown. We investigated the temporal expression of iNOS in a collagenase-induced ICH model of rats by RT-PCR, Western blot assay, and immunohistochemistry. Methods: ICH was induced to male rats by stereotaxic, intrastriatal administration of bacterial collagenase type IV. We investigated the expression of iNOS mRNA in the experimental ICH model rats. Rats were sacrificed at 0, 1, 3, 7 days after ICH. We also investigated the induction of iNOS in ICH on protein level by both immunohistochemistry and Western blot assay. Results: The induction of iNOS mRNA and protein was maximal at 1 day after ICH, then started to decrease at 3 days, and nearly normalized at 7 days after ICH. Immunoreactivity of iNOS in the brain was strongly expressed at 1 day after ICH. Conclusion: This novel results show that iNOS expression is maximal 1 day after ICH. There is the possibility that iNOS might be one of the pathologic mediators of ICH, and further study on its role in ICH is needed.
