To elucidate the cytotoxicity and antioxidative effect of Portulaca oleracea L. (PO) extract on copper (Cu), an allergic contact dermatitis(ACD)-induced agent, cell viability were analysed by XTT assay after the treatment of cupric sulfate (CuSO4) to cultured human skin fibroblasts (Detroit 551). And also, the effects of vitamin E and PO extract on CuSO4-induced cytotoxicity were measured after Detroit 551 cells were pretreated with vitamin E or PO extract for 2 hours. In the antioxidative effects of PO extract were measured by superoxide dismutase (SOD)-like activity, the inhibitory activity of lipid peroxidation (LP) and lactate dehydrogenase (LDH) activity. In this study, CuSO4 significantly decreased cell viability in dose-dependently compared with control, and the XTT50 value was calculated at 59.8 μM of CuSO4. While, vitamin E effectively prevented CuSO4-induced cytotoxicity and also, PO extract significantly increased cell viability compared with CuSO4-treated group. In the antioxidative effect, PO extract showed the inhibitory activity of LP and the decrease of LDH activity as well as SOD-like activity. From these results, it is suggested that the PO extract effectively prevented the cytotoxicity induced by an ACD-induced agent, CuSO4 via antioxidative effect. Conclusively, plant extract such as PO may be a putative resources as an alternative antioxidant for the prevention or diminution of ACD induced by allergen of heavy metals correlated with oxidative stress.
