This study was carried out to investigate the location of hair bulge area and to establish the basic technology for hair follicle stem cell culture. Hair root samples were obtained from plastic surgery as some surgical therapy of hair follicles, and then the putative hair follicle stem cells were cultured in Dulbecco’s Modified Eagle’s Medium and Ham’s F 12 Medium containing 10% fetal bovine serum, 10 ng/mL epidermal growth factor and 5 mg/mL insulin. Using hoechst dye staining,we confirmed that there were a number of cells in the bulge area of hair follicles. And the putative hair follicle stem cells were outgrown from the bulge area and succeeded until early four days. Therefore, further study will be continued to develop culture environment extending culture duration.
