To clarify the cytotoxicity induced by lead acetate (LA) and the protective effect of Elscholtziae splendense (ES) extract on the LA-induced cytotoxicity, cultured glioma cells (C6 glioma) were treated with or without LA or ES extract. In this study, LA significantly decreased the cell viability. While, vitamin E protected LA-induced cytotoxicity by the increase in cell viability which was decreased by LA. In the protective effect of ES extract on LA-induced cytotoxicity, ES extract prevented LA-induced cytotoxicity by a significant increase in cell viability, and showed the DPPH-radical scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory activity of lactate dehydrogenase (LDH). From these results, it is suggested that the cytotoxicity of LA is involved in oxidative stress, and ES extract prevented LA-induced cytotoxicity effectively. And also, it showed the antioxidative effects such as DPPH-radical scavenging activity, SODlike activity and inhibitory activity of LDH. Conclusively, the plant extract like ES may be a putative resources for the beauty by the diminution or blocking the cytotoxicity or oxidative stress mediated by heavy metals or chemicals which are components of hair dye or cosmetics.
