This study was undertaken to analyze the degree of hair damage after measuring activation energy from hair protein isotherm. The virgin hair were damaged by permanent wave after dyeing. The cuticles were observed with scanning electron microscope (SEM) to examine the hair damage. According to the observation of hair cuticle, the cuticles of the virgin hair were regular, close to each other and smooth, but the cuticles of the permanent wave hair after dyeing were shifted and shed. To evaluate the morphological damage of the virgin and permanent wave hair after dyeing observed with SEM image, the level of damage were classified into 5 levels, and evaluated by ten instructors of hairdressing. There was significant difference in hair damage. The hair damage of the permanent wave hair after dyeing (3.70±0.48) was higher than that of the virgin hair (1.60±0.52), but the tensile strength of the virgin hair (148.3±3.16 g) was higher than that of the permanent wave hair after dyeing (126.1±6.28 g) (p<0.05). The protein loss according to time sequence was analyzed quantitatively by the Lowry method after immersion in aqueous solution at 25℃ (298 K), 40℃ (313 K), and 70℃ (343 K), respectively. The protein loss was increased as time passed and increased with temperature elevation in aqueous solution. The amount of the protein loss was more in the permanent wave hair after dyeing than the virgin hair. On the contrary, the estimated activation energy values for protein loss was opposite, the activation energy of the virgin hair (74.9±3.66 kJ/mol) was higher than that of the permanent wave hair after dyeing (39.4±3.88 kJ/mol) (p<0.05). In conclusion, it is suggested that the activation energy from hair protein isotherm can be highly useful as degree of hair damage.
