In order to obtain an essential oil, a physiological activating substance, from the by-product of pine nut cone, in this research, steam distillation extraction(SDE) method and organic solvent extraction method by using ethanol, methanol, n-hexane and mixed solution were used. The extraction yields from each method were calculated and the contained compounds and their antibacterial activities were studied as following; The extraction from the mixed solution of ethanol and n-hexane (7:3) showed the highest yields of essential oil as 14.16% and the yield of ethanol extraction was 9.23%, from methanol extraction; 4.84%, from n-hexane extraction; 0.82%, and from SDE method; 0.4%, respectively. The essential oil from SDE method contained total 18 kinds of terpenoid and among them α-pinene(10. 58%) and D-limonene(9.84%) were analyzed as substances of biggest amount. In case of the extraction from organic solution, the results were a little different. The essential oil from ethanol extraction contained 9 terpenoids and among them 1-pheanthrenecarboxaldehyde(3.77%) and 1-phenanthrenemethanol(4.09%) were the compounds of biggest amount. The essential oil from the methanol extraction contained 9 terpenoids and 1-phenanthrenemethanol(6.69%), 1-phenanthrenecarboxaldehyde(6.40%) and 1-phenanthrenecarboxylic acid(5.77%) were obtained as main substances. On the other hand, total 13 kinds of terpenoids were analyzed from the essential oil from n-hexane extraction and 1-phenanthrenecarboxaldehyde (8.04%) and 1-phenanthrenecarboxylic acid (7%) were main compounds. In case of the essential oil from the mixed solution of ethanol and n-hexane(7:3), total 11 terpenoids were obtained and 1,4-methanoazulene(7.61%) and 1-phenanthrenecarboxaldehyde(7.62%) showed bigger portions than others. Moreover, the inhibition zone tests of the essential oil were also carried out in order to observe if the oil has antibacterial activity effect. As results, the antibacterial effect was confirmed against gram positive bacteria, such as Listeria monocytogenes and Staphylococcus aureus, even though there was no effect on gram negative bacteria like Eschericia coli and Salmonella typhimurium.
